Major Objectives

To teach students at the pre- and post-doctoral the three major
aspects of optical imaging:

(i) basic and advanced microscopy
(ii) imaging living specimens in 3-D
(iii) using F-techniques to probe cellular dynamics.



The specific topics taught will include; optical contrasting techniques, basics of video microscopy, image acquistion and processing, Confocal Microscopy, Multi-photon Microscopy, Selective Plane Illumination Microscopy (SPIM), and the F-techniques; Fluorescence Correlation Spectorscopy (FCS), Fluorescence Lifetime Imaging Microscopy (FLIM), Forster Resonance Energy Transfer (FRET), and Fluorescence Recovery After Photobleaching (FRAP).

 

At whom is it aimed; why is it important to teach these methods/systems/techniques?

The course is aimed at researchers working in the life sciences who currently use or wish to use basic and advanced optical techniques. In particular, scientists who are interested in observing live specimens in 3-D and measuring cellular dynamics. The participant will have a degree in the natural and/or life sciences and will be pursuing research in molecular, cell or developmental biology. He/she will have access to modern light microscopy equipment and computers but needs to understand the basic principles for his/her research in biology.

 

Recent developments in light microscopy, in particular in video microscopy, fluorescence microscopy, confocal microscopy as well as the continued development of GFP/fluorescent proteins had a dramatic impact on the life sciences. Numerous investigators use these techniques to address questions of fundamental biological importance and many more ask themselves how these can be used in their own research. The number of instruments as well as the number of their variants has increased and it is more complicated than ever to understand their relevance for the individual investigator. Two topics of great current importance are 3-D imaging and the F-techniques. By participating in the proposed course the student will gain experience of 3-D imaging and F-techniques allowing them to apply these techniques to their own research.
