About the Practical Course
Optical microscopy is in a state of explosive development. This is partly due to new technical advances, such as the use of photoproteins, computing and novel optics and partly to a shift of innovation from a few specialists working within manufacturing companies to a wider community of scientist users.

This EMBO course has been running for eight years. It starts with three lectures giving an overview of the diversity of microscopes and basic wave and ray optics, with students working in groups of three or less, each with their own research grade microscope.
Although there will be only 16 students, there will be a larger number of teachers and intensive practical instruction will be given, initially in setting up for Kohler illumination, phase contrast and DIC, cleaning lenses and alignment of mercury lamps.

At the same time, there will be demonstrations on Abbe theory, how to take apart and understand a microscope and quantitative interference microscopy.
Basic optical instruction will be continued in the second part of the course, with students performing optical bench experiments themselves on scanning systems and the effects of apertures.
The second week of the course will be organised with rotations of practical work, using the latest high-end microscope systems, including TIRF, confocal, multiphoton single spot and MP multispot systems and advanced cameras. More than 2.5 million euros worth of capital equipment is lent to the course by the principal manufacturers for this purpose.

There will be three hour-long lectures per day, ranging from the basics of detectors and light sources to the latest advances in nonlinear optics. Particular emphasis will be given to practical issues such as the imaging of living cells.
At the end of the course there will be the traditional presentation of the ‘Plymouth Glass Jellyfish Trophy’ to the student handing in the best answers to numerical questions on microscope optics (normally discussed in the local pub). |
